Fasciola hepatica is the mammalian liver parasite and it produces the result of poor welfare and economic losses in ruminants. While the number of eggs stool is the most common test used for diagnosis, it does not indicate a migration of the adult stage. serological techniques improve the sensitivity at all stages of liver fluke infection. The purpose of this study was to compare four commercially available ELISA test for the diagnosis of F. hepatica.
To this end, we tested three types of samples; (I) is known F. hepatica Status sera from experimental infection for the comparison of sensitivity and specificity, (ii) sera from pre- and post-flukicide-treated (albendazole, closantel, nitroxynil and triclabendazole) beef cattle seropositive for contrast difference before and after treatment, and (iii) samples of bulk tank milk samples from dairy cattle during periods of exposure to high and low F. hepatica to assess seasonal variation with four tests available. Samples were assayed using ELISA kit provided by four manufacturers (Ildana Biotech, IDEXX, Svanova, and Bio-X). Samples were analyzed simultaneously and in duplicate.In Ildana population control, IDEXX and Bio-X presented 100% sensitivity (Se) and specificity (Sp), Svanovir served Se and Sp of 59% from 96%.
In flukicide-treated beef, kit highlighted a decrease in antibody levels 90 days post-treatment in variable degrees. Finally, bulk milk showed a significant decrease in the value of ELISA between periods of high and low exposure to coincidence with all tests and Sp studied.Se found in this study, confirming that Ildana, IDEXX and Bio-X accurate for detecting exposure to F. hepatica in cattle Ireland. Svanovir Se and Sp in this population, suggesting that larger studies are needed to confirm the characteristics of these tests in the herd of Ireland.
In the use of post-treatment, Bio-X showed a consistent and significant reduction of the value of the ELISA in all treatment groups, which were shown to be a reliable tool to assess the effects of treatment on day 90 post-treatment. Finally, all tests showed to be a reliable tool for monitoring F. hepatica high season and low exposure, using bulk tank milk samples.

We compared the results of glutamic acid decarboxylase antibody test (GADAb) using radioimmunoassay (RIA) and enzyme-linked immunosorbent assay (ELISA) in subjects with age-onset diabetes mellitus type 1 specimen (T1D) .Serum collected from 1024 cases of Japan (426 laki- male and 598 female) in 2013. the average age of diagnosis was 7 years (0-18 years). Blood specimen obtained at a mean age of 13 years (2-22 years) .Among 628 subjects whose serum specimens collected within 5 years after diagnosis, positive GADAb level is 47.9% and 69.4% using a RIA using ELISA.
The subjects were divided into four groups according to their RIA and ELISA results for GADAb as follows: Gr I (RIA + / ELISA +), Gr II (RIA + / ELISA-), Gr III (RIA- / ELISA +) and Gr IV (RIA – / ELISA-). Clinical characteristics and genetic Gr Gr I and III were very similar in terms of age at diagnosis, male / female ratio, a relatively high positive rate for both IA-2AB and ZnT8Ab, and HLA genotype. Gr II only contains 5 patients and is characterized by a younger age at diagnosis, a low positive rate for both IA-2AB and ZnT8Ab and unique HLA genotype.

If the positive rate of both IA-2AB or ZnT8Ab or both are added to the results GADAb using RIA, autoimmune T1D percentage increased from 47.9% to 78.5% .the diagnosis of autoimmune time-onset T1D Japan GADAb increased when the results obtained using the new ELISA method , compared with RIA method previously used.
