HEV seroprevalence in blood donors in Turkey by two commercial total anti-HEV Ab ELISA kits.
Previous studies the prevalence of hepatitis E virus (HEV) in Turkey have shown high variability, leading to conflicting results. We aim to re-evaluate the HEV prevalence among blood donors in Turkey using Wantai (Beijing, China) and Dia.Pro (Milan, Italy) Total anti-HEV antibody (Ab) enzyme-linked immunosorbent assay (ELISA) kit and compare their performance and to investigate the presence of HEV RNA in blood donors. Total serum anti-HEV antibody is determined in a total of 2011 samples of voluntary blood donations collected from various regions of Turkey (807 from Ankara, 243 of Kayseri, 284 of İzmir, 200 of Malatya, Kahramanmaras 200, and 277 of Van). HEV RNA was evaluated by real-time polymerase chain reaction in a total of 272 anti-HEV seropositive samples.
A country-wide HEV seroprevalence was calculated as 11.5% (Dia.Pro) and 12.2% (Wantai) with seropositivity rate of 12.0% -12.5% in Ankara, 7.4% -8.2% in Kayseri, 14.5% -15.5% in Malatya, 8.1% -8.8% in Izmir, 15.0% -16.0% in Kahramanmaras, and 12.6% -13.4% in Van by Dia.Pro and Wantai kit, respectively. Ab lowest concentration detected was 0.16 and 0.14 units / mL WHO, for Dia.
Pro and test Wantai, respectively, showed no significant differences between the tests. HEV RNA was not detected in samples of anti-HEV seropositivity. Compared with previous studies, HEV shown to have a higher overall seroprevalence in Turkey. Despite its limitations, this study is the most comprehensive study of HEV seroprevalence in Turkey conducted by two different commercial ELISA test with high sensitivity so far. Further investigation is needed to determine the genotypes of HEV in Turkey.
Comparison of Two Commercial ELISA Kit for Detection of Anti-Dengue Dengue IgM for routine diagnosis in Laos.
The endemicity of dengue virus (DENV) infection remains a major public health problem in Lao PDR. In this study, we compared two commercial anti-dengue IgM ELISA kit, Panbio® Dengue IgM Capture ELISA (Panbio Kit, Alere, Waltham, MA, USA) and DEN DetectTM MAC-ELISA (InBios kit, InBios International, Inc., Seattle , WA, USA), in the context of the diagnosis of patients hospitalized with dengue clinical presentation. Two panels of paired blood samples tested.
A panel composed of 54 dengue patients confirmed (by DENV real-time RT-PCR) and 11 patients with non-dengue hemorrhagic fever (another infection confirmed by appropriate PCR). Panel B included 74 patients who were randomly selected from consecutive patients hospitalized Mahosot in 2008 with suspected dengue fever according to WHO criteria.
Results of panel A shows a significantly better sensitivity for Panbio kit (64.8%; 95% CI: 50.6 to 77.3%) compared InBios kit (18.5%; 95% CI: 9.3 to 31 , 4%) when testing sera entry. Increased sensitivity to both the kit when combining the results of the entrance and the sera recovered. The results obtained from the panel B in accordance with a fair agreement (κ = 0.29) between the two kits when testing a single sample entry, and deal moderate (κ = 0.5) when combining the results of the entrance and the sera recovered.
