Lyme disease (LD), caused by infection with Borrelia burgdorferi, is a tick-borne infection that is most common in many regions of Eurasia. detection of antibodies are laboratory tests are most often used, supports a two-step algorithm serodiagnostic; immunoenzymatic detection of antibodies to C6 has proven to perform similarly to the two-step standard workflow.
The purpose of this study is the evaluation of the performance of the C6 Lyme ELISA kit in comparison with a standard two-step algorithm in three laboratories located in the northeast region of Italy that is destined for areas with different epidemiological LD. A total of 804 samples tested, the 695 provides results in line between C6 and workflow testing routine (564 negative, positive 131).
Wherever available, the clinical presentation and additional laboratory tests are analyzed to resolve the differences. Methods based on C6 demonstrated good concordance with standard two-step algorithm (Cohen κ = 0.619), however, the distribution of difference seems to point towards a slightly lower specificity of the test C6, which is supported by the literature and could have an impact on patient management. C6 ELISA, therefore, not a stand-alone test that ideal; However, if it is integrated into a two-step algorithm, may be instrumental in achieving a sensitive, specific laboratory diagnosis of LD.
Development of Direct Competitive ELISA Kit for Detecting Deoxynivalenol Contamination in Wheat.
This research was conducted to develop a competitive enzyme-linked immunosorbent assay (dcELISA) direct assembly kit for detecting deoxynivalenol (DON) in food and feed grains. Based on the preparation of monoclonal antibody anti-DON, we established a standard curve with dcELISA and optimized detection conditions.
Performance kit was evaluated in comparison to high-performance liquid chromatography (HPLC). The minimum detection limit of the kit DON with 0.62 ng / mL, the linear range is 1.0 to 113.24 ng / mL and a half maximal inhibitory concentration (IC50) was 6.61 ng / mL in buffer work; there is a limit of detection (LOD) of 62 ng / g, and the detection range is 100-11324 ng / g in the sample authentic farm. We checked out four samples of wheat bran, wheat flour, corn flour and corn for the recovery of DON. Average recovery was in the range of 77.1% to 107.0%, and relative standard deviation (RSD) ranged from 4.2% to 11.9%.
In addition, the kit has the advantages of high specificity, good stability, long life effective and overlooked sample matrix interference. Finally, a sample of grain from farms in the six provinces of Henan, Anhui, Hebei, Shandong, Jiangsu and Gansu in China are analyzed with the kit. A total of 30 samples were randomly checked (five samples in each province), and the results are in good agreement with standard HPLC method. These tests showed that the kit dcELISA has good performance and meet the relevant technical requirements, and has the characteristics of accuracy, reliability, convenience and high-throughput screening for the detection of DON. Therefore, advanced kit is suitable for rapid screening of DON in the product being marketed.
