Preparation of ELISA and Lateral Flow Kits for rapid Diagnosis of Mycoplasma gallisepticum in Poultry
Lupi is a parasite Onchocerca emerging zoonosis of dogs, endemic to the southwestern United States and regions of the Old World. Currently, there are no specific serological diagnostic tests capable of detecting infection Lupi O .. Recent literature has shown that the heartworm antigen tests are commercially available, although highly sensitive, may cross-react with other filarid infection by nematode. No information on potential cross-reactivity of the test in the serum of infected dogs O. Lupi.
Our aim was to assess the serum samples of dogs naturally infected with O. Lupi potential cross-reactivity before and after heat treatment using a commercial ELISA kit heartworm. We obtained sera from 23 dogs naturally infected with O. Lupi. This dog presented with eye disease, and consulted to schedule either surgical removal of nodules eye due to infection or enucleation. Samples were assayed in triplicate using DiroCHEK® Heartworm Antigen Test kit (Synbiotics Corporation, Zoetis, Kalamazoo, MI, USA) following the manufacturers protocol pre- and post-heat treatment. The sample was heated using dry heat block at 103 ° C for 10 minutes and then centrifuged at 1818 × g for 20 minutes.
From a total of 23 dogs, 19 (82.6%) have no antigen was detected in spite of the heat treatment, the three dogs tested positive before and after heat treatment, and a single dog turned positive after heat treatment. All three dogs were positive before and after heat treatment confirmedly coinfected with heart worm by a veterinarian who is responsible for this case, and we were unable to get the historical or follow up with a dog that turns positive after heat treatment only.
Our data indicate that infection with O. Lupi should not result in false-positives when using DiroCHEK® in dog serum, before or after heat treatment. Dogs with a clinical eye onchocercosis that antigen-positive in DiroCHEK® may be co-infected with D. immitis, and should be tested, including the evaluation of microfilariae in the blood and diagnostic imaging. If the heartworm infection is confirmed, the animal must be listed in the recommended treatment protocol in accordance with the guidelines of the American Heartworm Society or other local organizations.
Commercial fishing ELISA kit has limited capacity to detect the different fish species and their products.
Fish is the main food and sources of allergens, requires a declaration on food packaging, are often determined by using ELISA. More than 1,000 different fish species traded and consumed around the world, increasingly provided by aquaculture. Up to 3% of the general population at risk of allergic reactions are sometimes fatal to fish, requires avoiding stringent of these food commodities. The purpose of this study is to evaluate the capacity of the three commercially available ELISA assays to detect a variety of bone and cartilaginous fish and their products, it is important to ensure detection labeling.
The reliable and safe food from fish bones 57 ranged from 26% to 61%. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Cattle CP in the samples is then determined by comparing the OD of the samples to the standard curve.
